      Without a link to page where the problem is on your site it is quite hard for us to provide the best solution due to so many variables. We try to provide file names and line numbers for code changes and if changes have already been made then our advice may be incorrect.

      I placed module Latest News (Actueel) in position "user1"
      and Most Read Content (Populair) in position "user2"

      Instead I want to put the modules in user4 and user5, but this is not working

      I got a chance to see the modules and I am guessing there is an unclosed div tag somewhere as the main content was not all showing yet the 3 modules worked fine.

      Login moved to left when no modules showing in 4 and 6 on other pages. This is correct behaviour as the module positions are collapsible.
