changing layout and module posistions

i'm comfortable with CSS and HTML, and i'm new to the Joomla CMS...or a CMS altogether. I have a simple question.
Since this is my first foray into PHP: What files should i edit in order to change the positions of modules and layout? and should i just upload the edited layout to my server.
I'll be using dreamweaver, my favorite txt editor, and firefox extentions. I understand i can use the CMS to do this, in some way. But i'm not finding the information to do this. do i need better search terms?
